What is oral embryology?
The study of the prenatal development of the orofacial structures — including the face, jaws, teeth, and associated soft tissues — from the embryonic and fetal periods.
From which three germ layers do oral and dental tissues develop?
Ectoderm (enamel, oral epithelium), mesoderm/neural-crest-derived ectomesenchyme (dentin, pulp, cementum, PDL, bone), and endoderm (lining of posterior tongue/pharynx).
What is the role of neural crest cells (ectomesenchyme) in tooth development?
Cranial neural crest cells migrate into the facial region and form the ectomesenchyme that gives rise to dentin, pulp, cementum, periodontal ligament, and alveolar bone.
What is the stomodeum and when does it form?
The stomodeum (primitive mouth) is a midline ectodermal depression on the embryonic face that appears around the 4th week; it is the precursor of the oral cavity.
What is the buccopharyngeal (oropharyngeal) membrane and what is its fate?
A bilaminar membrane (ectoderm + endoderm, no mesoderm) at the floor of the stomodeum separating it from the foregut; it ruptures around day 24–26 to connect the oral cavity with the pharynx.
Which facial processes/prominences surround the stomodeum?
The single frontonasal process (above), the paired maxillary processes (laterally), and the paired mandibular processes (below).
From what do the medial and lateral nasal processes arise?
They form from the frontonasal process around the nasal placodes, which invaginate to become nasal pits.
What structures form from fusion of the medial nasal processes?
The intermaxillary segment, which gives rise to the philtrum of the upper lip, the premaxilla (anterior maxilla carrying the four incisors), and the primary palate.
What forms the primary palate and what does it contain?
The primary palate forms from the intermaxillary segment (fused medial nasal processes); it forms the premaxillary portion bearing the four maxillary incisors, anterior to the incisive foramen.
What forms the secondary palate?
The secondary palate forms from the two palatal shelves (lateral palatine processes) of the maxillary processes, which fuse in the midline; it forms the palate posterior to the incisive foramen.
What is the sequence of palatal shelf movement during palate development?
The palatal shelves initially grow vertically alongside the tongue, then elevate to a horizontal position above the tongue (around week 7–8), and fuse with each other, the nasal septum, and the primary palate.
What landmark marks the junction of the primary and secondary palates?
The incisive foramen — the boundary between the anterior premaxillary (primary) palate and the posterior (secondary) palate.
What is odontogenesis?
The process of tooth development, involving reciprocal interactions between the oral epithelium (ectoderm) and the underlying neural-crest-derived ectomesenchyme.
What are the morphological stages of tooth development?
Dental lamina (initiation), bud stage, cap stage, bell stage, followed by apposition (crown formation) and maturation.
What is the dental lamina?
A horseshoe-shaped band of thickened oral epithelium that grows into the ectomesenchyme around the 6th week and gives rise to the tooth germs (enamel organs).
What three components make up a tooth germ?
The enamel organ (from oral epithelium → enamel), the dental papilla (from ectomesenchyme → dentin and pulp), and the dental follicle/sac (from ectomesenchyme → cementum, PDL, alveolar bone).
What is the enamel knot and its significance?
A transient cluster of non-dividing cells in the enamel organ at the cap/bell stage that acts as a signaling center (secreting BMP, FGF, SHH, Wnt) controlling cusp formation and tooth shape.
What are the four cell layers of the enamel organ in the bell stage?
Outer enamel epithelium, stellate reticulum, stratum intermedium, and inner enamel epithelium (which differentiates into ameloblasts).
What is the function of the stratum intermedium?
A layer of flattened cells between the stellate reticulum and inner enamel epithelium that supports ameloblast function (high alkaline phosphatase activity) during enamel formation.
What is Hertwig's epithelial root sheath (HERS)?
A double layer of inner and outer enamel epithelium that extends apically (without stellate reticulum) to map out root shape and induce root dentin formation.
What are the epithelial rests of Malassez?
Remnants of the fragmented Hertwig's epithelial root sheath that persist in the periodontal ligament; they can give rise to periapical (radicular) cysts.
What is amelogenesis and what cells perform it?
Amelogenesis is the formation of enamel by ameloblasts, which differentiate from the inner enamel epithelium and secrete the enamel matrix.
What is the chemical composition of mature enamel?
About 96% inorganic (hydroxyapatite), and ~4% organic material and water — making it the hardest, most highly mineralized tissue in the body.
What are the two phases of amelogenesis?
The secretory phase (ameloblasts lay down full-thickness partially mineralized matrix via Tomes' processes) and the maturation phase (mineral content increases and organic matrix/water are removed).
What is the basic structural unit of enamel?
The enamel rod (prism), a keyhole-shaped column of hydroxyapatite crystals running from the dentinoenamel junction toward the tooth surface.
